Methodology

Polymerase Chain Reaction (PCR)

Specimen Requirements

Specimens must be collected with Cobas PCR Collection Kit. Only swabs contained in Cobas PCR Collection Kit may be used to collect patient specimens. Collect specimens as follows:

 

Specimen source is required.

 

Specimen Type: Conjunctiva (eye)

Container/Tube: Cobas PCR Collection Kit

Specimen Volume: Swab

Collection Instructions:

1. Apply a topical, ophthalmological proparacaine-based anesthetic solution to eye or eyes (optional). Patient should not have had fluorescein drops administered.

2. Using small swab, thoroughly swab inner surface of lower, then upper eyelid. If specimen is taken from both eyes, swab less affected eye first to avoid further contamination.

3. Immediately place swab into transport tube provided in collection kit.

4. Cap tube securely.

5. Label tube with patient’s name (first and last), date and actual time of collection, and type of specimen.

Specimen Type: Endocervix or vaginia

Specimen Volume: Swab

Collection Instructions:

1. Insert dry swab 2" into the vagina or 1 to 1-1/2 cm into endocervical canal and rotate vigorously for 30 seconds. 
2. Place swab into transport tube provided in collection kit and break the score line.

3. Cap tube securely.

4. Label tube with patient’s name (first and last), date and actual time of collection, and type of specimen.

Collection Instructions:

1. Collect specimen using swab provided.

2. Pass swab beyond anal sphincter.

3. Rotate swab in rectum.

4. Place swab into transport tube.

5. Label tube with patient’s name (first and last), date of birth, date and actual time of collection, and type of specimen.

Container/Tube: Cobas PCR Collection Kit

Specimen Volume: Swab

Collection Instructions:

1. Collect specimen using swab provided.

2. Tilt patient’s head back to assist in opening of mouth as wide as possible.

3. Depress tongue with tongue depressor so swab doesn’t touch oral mucosa or tongue.

4. In 1 continuous motion:

a. Swab 1 tonsillar area up then down.

b. Move to back of throat as far down as possible and swab there.

c. Move to other tonsillar area and swab.

d. Swab behind uvula and remove swab.

5. Place swab into transport tube.

6. Label tube with patient’s name (first and last), date of birth, date and actual time of collection, and type of specimen.

Collection Instructions:

1. Clean-catch, mid-stream specimen is not acceptable.

2. Patient should be instructed not to urinate for 1 hour prior to sampling. 
3. Label container with patient’s name (first and last), date and actual time of collection, and type of specimen.
