Test Code LAB2616 Coagulation Factor IX Activity Assay, Plasma

Additional Codes

Test Name in EPIC EPIC Test Code Mnemonic
FACTOR IX ASSAY LAB2616 FAC9

Useful For/Utility

Diagnosing deficiencies, particularly hemophilia B (Christmas disease).  Assessing the impact of liver disease on hemostasis.   Investigation of a prolonged activated partial thromboplastin time.

Methodology

Electromechanical Clot Detection

Clinical Information

Factor IX is a vitamin K-dependent serine protease synthesized in the liver and participates in the intrinsic coagulation pathway. Its biological half-life is 18 to 24 hours.  Congenital deficiency inherited as an X-linked recessive bleeding disorder (hemophilia B).   Acquired deficiency associated with liver disease, vitamin K deficiency, warfarin therapy and inhibitors (rare).

Specimen Requirements

Specimen Type: Platelet-poor plasma

Container/Tube:

Fill Blue top tube to the etched line located just below the cap of the tube.

Preferred: Full Light-blue top (3.2% sodium citrate)

Acceptable: Full Sodium Citrate (BD® or Greiner tubes)

Specimen Volume: 1 mL

Specimen Minimum Volume: 0.5 mL

Collection Instructions:

1. Do not spin in refrigerated centrifuge.

2. Unacceptable specimens: Specimens that are short-draws, over-draws, clotted or hemolyzed may yield incorrect results.

3. If sending from a site spin down, remove top 2/3 of plasma, and spin plasma again in a polypropylene (cloudy plastic) vial.

4. Aliquot top 3/4 and place platelet-poor plasma in a polypropylene (cloudy plastic) vial.

5. Glass vials are not acceptable for processing/transport/storage.

Additional Information:

1. Double-centrifuged specimens are critical for accurate results as platelet contamination may cause spurious results.
2. If there is a delay in transport of > 4 hours, send specimen frozen.

Specimen Transport Temperature

Frozen 15 days at -20° C or 1 month at -80° C OK/Ambient 4 hours at 20° ± 5° C OK/Refrigerate NO
