Useful For/Utility

To enumerate an individual's major lymphocyte subsets - T, B and Natural Killer (NK) Cells.  Includes percent cells and absolute counts for CD3 (T-cells), CD4 (helper cells), CD8 (suppressor cells), CD19 (B-cells), CD20 (B Cells) and CD3-CD56+CD16+ (Natural Killer Cells), and Helper/Suppressor ratio.

Methodology

Immunophenotyping by Flow Cytometry

 

Purpose: To enumerate an individual's major lymphocyte subsets - T, B and Natural Killer (NK) Cells.

 

Includes percent cells and absolute counts for CD3 (T-cells), CD4 (helper cells), CD8 (suppressor cells), CD19 (B-cells), CD20 (B Cells) and CD3-CD56+CD16+ (Natural Killer Cells), and Helper/Suppressor ratio.

Clinical Information

CD3+, CD4+, CD8+ and/or CD19+ lymphocyte percentages and absolute counts may be used as aids to evaluate immune competency, underlying known or unknown disease states, and to monitor lymphocyte
levels following organ transplantation.
Identification of abnormal levels of CD4+ immunodeficiency and corresponding CD4+/CD8+ ratios might also aid in the diagnosis and/or prognosis of immunodeficiency disease.   CD4/CD8 ratios in conjunction with CD4+ lymphocyte cell numbers have been the most widely used laboratory parameters for
evaluation of AIDS-related complex and AIDS.

Specimen Requirements

Specimen must be sent to SMDC Main Laboratory the day of collection or ASAP thereafter. Testing must be completed within 48 hours of draw.

Specimen Type: Peripheral whole blood

Container/Tube: 4 mL Lavender top (EDTA)

Specimen Volume: Full tube

Specimen Minimum Volume: 1.0 mL

Additional Information: Specimen cannot be frozen.

 

Collection Instructions:

  1. For serial monitoring, it is recommended that specimen draws be performed at the same time of day.
  2. Testing must be completed within 48 hours of specimen collection.
  3. It is recommended that specimens arrive within 24 hours of draw. Draw and package specimen as close to shipping time as possible.
  4. Forward promptly to Flow Cytometry Lab at ambient temperature.
  5. Send specimen in original tube. Do not aliquot.
