Methodology
Microscopic Examination of Stained Buffy Coat Slides
Specimen Requirements
EDTA whole blood and smears are required.
Specimen Type: Whole blood
Container/Tube: Lavender top (EDTA)
Specimen Minimum Volume: 1 mL
Additional Information:
1. Include a copy of hemogram results.

Specimen Type: Smears
Container/Tube: Slide container
Specimen Volume: 6 smears
Collection Instructions:
1. Draw blood in a lavender-top (EDTA) tube
2. Prepare 2 peripheral smears (labeled as peripheral smears) and 4 buffy coat smears (labeled as buffy coat smears).
3. Prepare buffy coat smears as follows:
a. Spin down a small portion of EDTA whole blood.
b. Pipet buffy coat along with a small portion of plasma and make 6 smears.
c. Label these smears as buffy coat smears.

Specimen Transport Temperature
Use one 5 ml EDTA tube less than 4 hours old for optimum results. If specimens are transported to the lab for analysis, it is preferable to make slides from fresh blood to send.
