Methodology

Hemogram with Differential

Impedance, Direct Current (DC) detection method, and Particle Distribution Analysis (PDA).

Includes WBC, RBC, hemoglobin, hematocrit, platelets, RDW, MCV, MCH, and MCHC, and a differential (neutrophil, lymphocytes, monocytes, eosinophils, basophils, and Immature grans)

 

Anaplasma (Ehrlichia) Smear

Microscopic Examination of Stained Buffy Coat Slides

Reflex Information:

If WBC <4 or PLT <150, an anaplasma (Ehrlichia) smear will be performed.

Specimen Requirements

EDTA whole blood and smears are required.

Container/Tube: Lavender top (EDTA)

Specimen Minimum Volume: 1 mL

Additional Information:

1. Include a copy of hemogram results.

Collection Instructions:

1. Draw blood in a lavender-top (EDTA) tube

2. Prepare 6 peripheral smears (labeled as peripheral smears) and 6 buffy coat smears (labeled as buffy coat smears).

3. Prepare buffy coat smears as follows:

a. Spin down a small portion of EDTA whole blood.

b. Pipet buffy coat along with a small portion of plasma and make 6 smears.

c. Label these smears as buffy coat smears.

Specimen Transport Temperature

Ambient 24 hours/Refrigerate 48 hours OK/Frozen NO

If manual differential is required, slides must be made within 8 hours of collection.

Buffy Coat smears should be made within 4 hours if possible and no longer than 8 hours after collection.
